The Ki-67 Antibody In Tumour-cell Marker Studies

The Ki-67 protein is a member of the FHA (Forkhead Associated Domain) family of nuclear proteins, which are involved in DNA repair, transcriptional regulation and establishing/maintenance of cell cycle checkpoints. Ki-67 is expressed during all active phases of the cell cycle, but is downregulated during the G0 resting phase, making it an excellent cellular marker for proliferation studies. The Ki-67 protein was identified by J. Gerdes, et al. in 1983, using a prototype monoclonal Ki-67 antibody raised in mice immunized with Hodgkin lymphoma nuclei (cell line L428.) In 2000, J. Gerdes and T. Scholzen published a paper suggesting Ki-67 is specific and essential to cell proliferation, as it is restricted to the nucleus during interphase but relocates to the chromosome surface during mitosis.

Gerdes’ 2000 study used Ki-67 antibody preparations to demonstrate their application in the diagnosis of human breast, brain and prostate tumours. The Ki-67 labeling index, which is the fraction of Ki-67-positive tumour cells, often correlates with the progression of the disease.

In 2005 Gerdes, Scholzen, et al. published evidence suggesting that Ki-67 is also associated with ribosomal RNA transcription in proliferating and resting cells. Despite downregulation in the G0 phase, the Ki-67 antibody still detected the protein at sites involved with rRNA synthesis. In proliferating cells, pKi-67 was found at rRNA transcription sites during mitosis and interphase. Together with data from ChIP Ki-67 antibody assays, this strongly suggested Ki-67 is involved in early rRNA synthesis as well as cellular proliferation.
